Title: Elucidating the principles of antigen presentation during Mtb infection
Summary: T cell recognition of Mycobacterium tuberculosis (Mtb)-infected cells contributes to immunity to tuberculosis (TB), but the principles that govern presentation of Mtb antigens on MHC-I and MHC-II in human cells are incompletely understood. Overcoming this knowledge gap would provide new insights for TB vaccine development. We utilized mass spectrometry (MS) analysis to elucidate the MHC-I and MHC-II repertoire of Mtb-infected primary human macrophages and utilized a combination of bacterial genetics and host perturbations to mechanistically probe how presentation of Mtb-derived antigens occurs. In our recent work, we have further developed new techniques to systematically study bacterial protein secretion in vivo.
